MAGL inhibitor 4f (T-211) is a potent, brain-penetrant reversible monoacylglycerol (MAGL) inhibitor with IC50 of 6.2 nM and 13 nM for human and mouse MAGL respectively, with high selectivity over FAAH and ABHD6 (IC50 > 10,000 nM).
MAGL inhibitor 4f (T-211) (1 mg/kg) robustly increased 2-arachidonoylglycerol levels and decreased arachidonic acid levels in the mouse brain.
MAGL inhibitor 4f (T-211) (1 mg/kg) significantly decreased the cytokine and chemokine expression levels and suppressed neuronal cell loss in the hippocampi of kainic acid (KA)-injected mice.
MAGL inhibitor 4f (T-211) also ameliorated cognitive impairment in KA-injected mice.
