MIRO1 Reducer 3 (MR3, Miro1 Reducer) is first-in-class small molecule that allosterically disrupt mitochondrial Rho GTPase 1 (MIRO1)-BAX complex, binds C-GTPase domain of the MIRO1, blocks BAX activation and membrane destruction.
MR3 treatment significantly reduces cell viability across all glioma lines in a dose-dependent manner, does not affect non-glioma cancer or non-cancer cells.
MR3 prevented Antimycin A (AMA)-induced cell death in SNCA-A53T iDAs.
MR3 also rescues DA neuron loss in a Drosophila PD model expressing human SNCA-A53T in vivo.
MR3 (10 uM) altered BAX protein levels in glioma, markedly reduced the number of mitochondria-localized BAX puncta.
MR3 sensitizes glioma cells to ferroptosis and protects Parkinsonian neurons from apoptosis.
MIRO1 Reducer 3 (MR3, Miro1 Reducer) eliminates the Miro1 defect in PD fibroblasts, rescues Parkinson's neurodegeneration.
