MK-8153 (MK8153) is a potent, selective inhibitor of renal outer medullary potassium channel (ROMK, Kir1.1) with IC50 of 5 nM, 3,400-fold over hERG.
MK-8153 inhibits current through rat ROMK, rKir1.1/HEK293 cells in electrophysiological recordings with an IC50 value of 2.5 nM.
MK-8153 also shows no activity against related inward rectifying potassium channels (Kir2.1, Kir4.1, and Kir7.1 IC50s > 100 μM) as well as the cardiac channels Nav 1.5 and Cav 1.2 (>30 μM).
MK-8153 (1-3 mg/kg/day) caused a dose-dependent decrease in systolic blood pressure in aged SHRs.
