MRL-494 (MRL494) is an antimicrobial small molecule that inhibits assembly of outer membrane proteins (OMPs) by the β-barrel assembly machine (BAM complex), inhibits the insertion of OMPs into the OM by targeting BamA.
MRL-494 exhibited antimicrobial activity against a broad range of bacterial species, impairs biogenesis of OMPs and potentiates rifampicin.
MRL-494 impairs biogenesis of OMPs, BamAE470K provides resistance to MRL-494 and restores OMP biogenesis.
MRL-494 inhibits Gram-positive and -negative microbes through distinct mechanisms of action.
