MRV03-069 is a small molecule compound that potently and specifically inhibit the colibactin-activating peptidase ClbP (IC50=28 nM), inhibits colibactin-associated genotoxicity.

MRV03-069 is a small molecule compound that potently and specifically inhibit the colibactin-activating peptidase ClbP (IC50=28 nM), inhibits colibactin-associated genotoxicity.
MRV03-069 does not appear to interact with any of the other serine hydrolases.
MRV03-069 engages the ClbP binding pocket, forming a covalent bond with the catalytic serine.
MRV03-069 could reproduce the phenotypes observed in a clbP deletion mutant and block the genotoxic effects of colibactin on eukaryotic cells.
MRV03-069 also inhibits purified ZmaM with similar potency to ClbP in vitro, a ClbP homolog that is proposed to activate the antibiotic zwittermicin.
