MSU-43085 is a potent, orally bioavailable inhibitor of Mycobacterium tuberculosis (Mtb, EC50=120 nM) and M. abscessus survival, targets MmpL3.
MSU-43085 is ~10-fold higher in potency than the clinically studied MmpL3 inhibitor SQ109 but ~3- to 5-fold lower than NITD-304 and NITD-349.
MSU-43085 exhibits potent growth inhibition of intracellular Mtb with EC50 of 134 nM.
MSU-43085 is potent against NTM species MAB and MAC with MICs of 2.9 and 23 µM against MAB and MAC, respectively.
MSU-43085 is active against drug-resistant Mtb strains.
MSU-43085 (200 mg/kg) is efficacious in an acute model of Mtb infection.
