MTX-3937 is a small-molecule Siglec-9 inhibitor with binding KD of 3.15 uM in BLI assay, inhibits phosphorylation of Siglec-9 and downstream SHP1 and SHP2.
MTX-3937 shows similar binding affinities towards Siglec-7 and Siglec-9.
MTX-3937 suppresses Siglec-9-induced inhibitory signaling and promotes NK cell functions, inhibits NK cell apoptosis.
MTX-3937 increases NK cell homeostasis, improves the functions and homeostasis of NK cells from patients with HCC or HBV.
MTX-3937 remarkably improves NK cell-mediated anti-tumor efficacy against liver cancer in vivo.
