MU262 is a specific small molecule inhibitor of endonuclease MUS81 with IC50 of 0.87 uM in biochemical nuclease assay, inhibits MUS81-mediated DNA repair pathways in cells.
MU262 has no negative effect on NHEJ, does not broadly suppress DNA repair but impair only HR/BIR pathways.
MU262 decreases BIR to the same extent as the siRNA-mediated depletion of MUS81, with an IC50 of 1.53 uM, also significantly reduces the efficiency of HR repair with IC50 of 0.5-1 uM.
MU262 likely impairs the MUS81 enzymatic activity through an alternative, DNA binding-independent mechanism.
MU262 significantly increases micronuclei formation in U2OS cells, similarly to the effect of MUS81 depletion.
MU262 induces persistent DNA damage leading to reduced cell survival, synergizes with cisplatin to reduce cancer cell viability.
