Biological Activity
MWAC-3475 is a potent small molecule Zika virus (ZIKV) inhibitor targeting the viral protein NS4B with EC50 of 0.63 uM in CPE-based assays.
MWAC-3475 shows no activity against other flaviviruses, including DENV, Yellow fever virus (YFV), Japanese encephalitis virus (JEV), and West Nile virus (WNV).
MWAC-3475 induces significant, concentration-dependent, reduction in progeny virus titers, > 1000-fold reduction at 5 µM and reaching a 4.2-log reduction (or 1.78 x 10 4-fold) at 20 µM in ZIKV- infected Vero 76 cells.
MWAC-3475 inhibits viral replication in the middle stage and require de novo protein synthesis.
