Mtb GuaB2 inhibitor G9 is a potent, specific inhibitor of Mycobacterium tuberculosis inosine 5'-monophosphate dehydrogenase (Mtb GuaB2) with IC50 of 0.94 nM, and potent MIC against virulent Mtb H37Rv (IC50=58 nM).
G9 shows high selectivity for bacterial GuaB over the human homology hIMPDH2 (hIMPDH2 IC50 >50 uM).
G9 is efficacious against Mycobacterium tuberculosis as a single agent and in combination with other antibiotics in the chronic C57BL/6, BALB/c, and C3HeB/FeJ mouse models of lung infection.
