MyD88 inhibitor 3g is a potent, TIR domain-selective and orally bioavailable MyD88 inhibitor, binds TIR domain (SPR KD=28.5 uM) but not the DD domain, disrupts MyD88 dimerization.
MyD88 inhibitor 3g significantly suppressed IL-6 secretion from J774A.1 cells (IC50 = 1.32 μM) and THP-1 cells (IC50 = 0.75 μM). MyD88 inhibitor 3g inhibited the activation of the NF-κB and MAPK pathways stimulated by LPS.
MyD88 inhibitor 3g exhibited anti-inflammatory function by binding to the TIR domain instead of the DD domain.
MyD88 inhibitor 3g demonstrated significant anti-inflammatory efficacy and offered substantial protection against LPS and CLP-induced ALI.
