MyD88 inhibitor A5S is a specific small molecule MyD88 inhibitor with SPR Kd of 15 uM, exerts anti-inflammatory effect through the NF-κB/MAPK pathway.
A5S reduces release of the LPS-induced IL-6 and TNF-α in J774A.1 cells with IC50 of 0.24 uM and 3.28 μM, respectively.
A5S restores IκB-α levels and suppressed the phosphorylation of p65.
A5S interferes with MyD88 homologous dimerization and inhibits MAPK pathway activation.
A5S protects mice from LPS-induced and sepsis-induced acute lung injury (ALI) mouse models.
A5S is a MyD88 inhibitor of not affecting the expression of IFN-β in acquired immunity in suppressing excessive inflammatory response.
