NAB2 (Nedd4 inhibitor NAB2) is a specific small molecule inhibitor of E3 ubiquitin ligase Nedd4 with binding KD of 42 nM.
NAB2 rescues α-synuclein-associated trafficking defects associated with early onset Parkinson's disease in a Nedd4-dependent manner.
NAB2 binds to Dedd4 side chain and does not alter its conformation or ubiquitination kinetics in vitro, does not alter its co-localization with trafficking organelles.
