NB-360 is a potent, selective and brain penetrable dual BACE1/BACE2 inhibitor with IC50 of 5 nM/6 nM respectively.
NB-360 does not inhibit pepsin or the endosomal/lysosomal cathepsins D and E (IC50 > 250 μM).
NB-360 is a potent inhibitor of Aβ40 release from human APP-overexpressing CHO cells, with an IC50 of 3 nM in wtAPP CHO cells and 33 nM in SweAPP CHO cells.
NB-360 potently inhibits the release of Aβ42 (IC50: 3 nM) and sAPPβ (IC50: 4 nM) in wild-type APP CHO cells.
NB-360 is sufficient to completely block amyloid-β deposition in an APP transgenic mouse model.
NB-360 displays dose-dependent appearance of irreversibly depigmented hair, induces hair depigmentation by inhibiting PMEL17 processing in mice.
