NDI-091143 (NDI091143) is a potent, small-molecule inhibitor of human ATP-citrate lyase (ACLY, ACL) with Ki of 7.0 nM and IC50 of 2.1 nM in the ADP-Glo assays.
NDI-091143 is located in an allosteric, mostly hydrophobic cavity next to the citrate-binding site, and requires extensive conformational changes in the enzyme that indirectly disrupt citrate binding.
