NH300094 is a potent antagonist of 5-HT receptors and dopamine receptors with IC50 of 0.47 nM, 1.04 nM, 11.71 nM and 31.55 nM for 5-HT2A, D2L, D2S and D3 receptors, respectively.
NH300094 is a triple antagonist of 5-HT2A receptor, dopamine D2 receptor and dopamine D3 receptor.
NH300094 shows high inverse agonist activity of 5-HT1BR (EC50 = 28.36 ± 12.52 nM) and antagonistic activities of 5-HT1AR (IC50 = 85.59 ± 61.53 nM) and D3R (IC50 = 31.55 ± 23.08 nM).
NH300094 (0.01, 0.03, 0.1 and 0.3 mg/kg, oral) resulted in a dose-dependent inhibition of MK-801-induced hyperactivity in male ICR mice with an ED50 of approximately 0.07 mg/kg and Minimum effective dose (MED) of 0.03 mg/kg.
NH300094 shows powerful efficacy in positive, negative, and cognitive impairment animal models.
