NN-429 (NN429) is a potent, selective second-generation inhibitor of HDAC6 with IC50 of 3.2 nM, >312-fold selectivityt over all HDAC isoforms.
NN-429 is an improved analog of older generation HDAC6 inhibitor molecules KT-531 and NN-390, with higher in vitro selectivity, target potency and in vivo half-life.
NN-429 displays IC50 value of 28.8 nM in fluorescence polarization (FP) assays.
NN-429 shows selective cellular cytotoxicity in a spectrum of T-cell lymphoma malignancies.
NN-429 dose-dependently increases the acetylation of α-tubulin in acute myeloid leukemia (AML) cells (MV4-11) at as low as 0.1 uM.
NN-429 exhibits strong synergy with clinical agents cytarabine, doxorubicin, etoposide, and with SNS-032 in in cellular models of γδ T-NHL and NKTCL.
