No. 10-3 (PSF inhibitor No. 10-3) is a small molecule that inhibits RNA binding by PSF with IC50 of 2.2 pM (nhibiting the interaction of PSF with CTBP1-AS probe), decreases prostate cancer cell viability (22Rv1 cells IC50=1.2 uM).
No. 10-3 inhibited the PSF-mediated RNA splicing and the epigenetic pathway that promotes apoptosis and inhibits cell cycle in both breast and prostate cancer cells.
Pharmacologic inhibition of PSF function by No. 10-3 impaired tumor growth in a xenograft mouse model of treatment-resistant prostate cancer.
RNA-binding protein PSF functions as an epigenetic modifier by interacting with long noncoding RNAs and the corepressor complex. PSF also promotes RNA splicing events to enhance oncogenic signals.
