Biological Activity
OPC-33540 (OPC33540) is a potent, highly selective and competitive PDE3 inhibitor with IC50 of 0.32 nM and 1.5 nM for PDE3A and PDE3AB respectively.
OPC-33540 displays little to no activity against PDE1, PDE2, PDE4, PDE5, and PDE7.
OPC-33540 potentiates cyclic AMP accumulation concentration-dependently in both the absence and the presence of 3 nM prostaglandin E1 (PGE1) (doubling concentrations: 32.5 and 6.2 nM, respectively) in platelets.
OPC-33540 inhibits thrombin-induced platelet aggregation with IC50 of 27.8 nM.
OPC-33540 inhibits anti-platelet aggregation effect stimulated in the presence of 3 nM PGE1 with IC50 of 6.0 nM.
