PAD-PF2 is a potent inhibitor of peptidylarginine deiminases (PAD1-4) with IC50 of 42.7 nM for PAD4 mediated citrullination in the fluorescent quenching assay, inhibits all four catalytically active PAD1-4 with comparable potency in a Ca2+-dependent manner.
PAD-PF2 binds similarly to both PAD4 and PAD2.
PAD-PF2 showed > 80-fold selectivity against a diverse panel of receptors, enzymes, kinases, transporters, and ion channels.
PAD-PF2 inhibits ionomycin induced protein citrullination in human isolated neutrophils.
PAD-PF2 could fully inhibited CitH3 detected protein citrullination with IC50 of 1.0 uM, also able to fully inhibit F95 detected protein citrullination (IC50 = 1.5 uM).
