PAV-174 is a host protein-targeted antiviral compound that specifically interacts with an oxidized conformer of MIF (oxMIF), inhibits HSV-1 replication in a dose-dependent manner in HSV-1-infected Vero cells, both in a standard plaque assay and in an in-cell ELISA with an IC50 of 34 or 21 nM, respectively.

PAV-174 binds MIF at its multimerization interface.
PAV-174 reduces tau phosphorylation also in the absence of infection in an MIF- and dose-dependent manner.
PAV-174 inhibit oxMIF-induced tau phosphorylation in vitro and in 2-month-old male tgTau58/2 mice independent of infection.
