Biological Activity
PELI1 inhibitor BZ61 is a small molecule inhibitor of E3 ubiquitin ligase Pellino-1 (PELI1) with SPR Kd of 1.985 uM, suppresses the proliferation of both tyrosine kinase inhibitors (TKIs)-sensitive and TKI-resistant CML cells.
BZ61 treatment markedly reduced the expression of both Snail and STAT3 in K562 cells, suppressed K63-linked ubiquitination of Snail and STAT3.
BZ61 inhibited K562 cells proliferation in a dose-dependent manner with IC50 of 13.6 uM.
BZ61 markedly reduced the interaction between PELI1 and BCR-ABL1 in Co-IP assays, induced a pronounced decrease in BCR-ABL1 protein levels but not mRNA levels.
BZ61 suppressed human CML burden and progression in vivo.
