Peptide Targeting BamA-1 (PTB1) is a 12-amino acid macrocycle, multi-species BamA inhibitor, binds to purified BamA with SPR Kd of 50 pM, has MIC of 42 ug/mL (25 uM) against wild-type E. coli strain.
PTB1 exhibits MICs against wild-type Klebsiella pneumoniae and Enterobacter cloacae, ranging from 42 to 168 µg/mL (25–100 µM), shows no MICs against the non-fermenting Gram-negative pathogens Pseudomonas aeruginosa and Acinetobacter baumannii.
PTB1-1 reduces OMP levels, decreases activity of the OmpT enzyme that requires BAM for folding, and increases OM permeability.
PTB1 targets an extracellular binding site, and acts as a state-dependent antagonist which targets the closed state of BamA.
