PanK inhibitor MNS is a broad-spectrum antifungal, fungal-selective inhibitor of PanK with IC50 of 1.13 μM and 17.74 μM against CaCab1p and AfPanKp respectively, directly engages and inhibits PanK to block CoA production.
MNS exerts relatively potent antifungal activity against the C. albicans reference strain – SC5314 (CAB1/CAB1), with an MIC50 of 1.95-3.9 μM.
MNS does not inhibit human PanK1 at any concentration tested (IC50 > 400 μM).
MNS shows dose-dependent reductions in intracellular CoA concentrations in treated wild-type C. albicans cells.
MNS (0.1 mg/kg/day) has antifungal efficacy in a mouse model of disseminated Candida albicans infection.
