Phenylacetamide 2 (FFA2 allosteric agonist 2) is a potent, specific and allosteric agonist of FFA2 (GPR43) with cAMP IC50 of 0.48 and 0.66 uM for hFFA2 and mFFA2, respectively.
Phenylacetamide 2 is inactive against a panel of GPCRs that included FFA1, FFA3, GPR109A, growth hormone secretagogue receptor, endothelin type B receptor, CCR2, CXCR3, CXCR4, and CCR7 at 30 uM.
Phenylacetamide 2 is capable of activating both Gαq- and Gαi -coupled pathways for receptor function.
Phenylacetamide 2 inhibits lipolysis in 3T3L1 adipocytes in a concentration-dependent manner as measured by the reduction of glycerol production in adipocytes.
