Phox-I2 is a specific small molecule inhibitor of Rac1-p67phox interaction, binds to the Rac1 interactive site of p67phox specifically (Ki=150 nM) and interferes with Rac1-GTP interaction with p67phox.
Phox-I2 is active in suppressing ROS production in both human and murine neutrophils.
Phox-I2 is effective in inhibiting NOX2-mediated superoxide production dose-dependently in human and murine neutrophils without detectable toxicity.
Phox-I2 inhibitedcollagen-induced platelet aggregation without affecting the tail bleeding time, inhibited in vivo platelet adhesion/accumulation at the laser injury sites on the saphenous vein without affecting the time for complete cessation of blood loss.
