RAC1 inhibitor A41 is a potent, specific and reversible inhibitor of RAC proteins, competes with guanine nucleotide binding, inhibits RAC1-dependent membrane ruffle formation with IC50 of 2.5 nM.
A41 (10 uM) blocks RAC1 activation in EGF-stimulated NIH/3T3 fibroblasts.
A41 is not RAC-GEF dependent.
A41 (10 uM) reduced TRIO-induced activation of RAC2, which is closely related to RAC1, but had no effect on TRIO-induced RHOG activation.
A41 directly interacts with RAC1 (SPR KD = 30 nM), shows no affinity for RHOA and CDC42.
A41 inhibits RAC1 activity and oncogenic properties of TNBC cell line, decreases growth and invasiveness of TNBC cells in vitro.
A41 (25 mg/kg/day, i.p.) prevents metastasis in a mouse model of TNBC.
