Biological Activity
RANKL inhibitor 34 is a potent and selective RANKL/RANK inhibitor, displays high affinity for RANKL with SPR Kd of 614 nM, effectively inhibits RANKL-induced osteoclastogenesis and bone resorption.
RANKL inhibitor 34 displays >100-fold selectivity over TNF based on binding affinity.
RANKL inhibitor 34 (5.0 μM) even completely inhibited osteoclast formation.
RANKL inhibitor 34 effectively downregulates the c-fos/NFATc1 signaling in BMMs in a dose-dependent manner.
RANKL inhibitor 34 decreases the mRNA levels and the expression of nuclear receptor NFATc1 and c-fos that controlled the expression of bone resorption enzymes such as TRAP and cathepsin K.
