RBC8 is a specific small molecule inhibitor of GTPase Ral, inhibits both RalA and RalB activation and colony formation of Ral-dependent lines H2122 and H358 in soft agar with IC50 of 3.5 and 3.4 uM, respectively.
RBC8 acts specifically through the GDP-bound form of Ral proteins, RalAG23V or RalBG23V, mitigates inhibition of H2122 and H358 cell growth.
RBC8 shows selectivity for Ral relative to Ras or Rho and inhibits xenograft tumor growth similar to depletion of Ral by siRNA.
RBC8 also enhances the inhibitory effects of imatinib in K562, KCL22 and BaF3-P210 cells.
