Biological Activity
RK783 is a potent, selective inhibitor inhibitor of BMP type 1 receptor kinase (ACVR1), inhibits CVR1-Q207D, ACVR1-R206H, wild-type ACVR1, and other BMP type 1 receptor kinases with IC50 of 10-120 nM.
RK783 preferentially suppressed both basal and stimulated BMP-Smad1/5/9 signaling in vitro without affecting the signaling of Smad2/3.
RK783 specifically suppress BMP-Smad signaling and prevent HO formation in both the ACVR1-R206H and ACVR1-Q207D mouse efficacy models.
RK783 can also induce myofiber repair and inflammation infiltration.
