Biological Activity
RO7019009 is a small molecule γ‐secretase modulator, efficiently reduces the levels of Aβ42 (IC50=14 nM) and Aβ40 secreted by HEK293/sw cell, effectively lowers Aβ43 of all Aβ43‐generating mutants.
RO7019009 has IC50 values for brain‐derived human neuroglioma H4 (IC50 (Aβ42) = 5 nM) or mouse neuroblastoma N2a (IC50 (Aβ42) = 2 nM) cell lines overexpressing swAPP (H4/sw, N2a/sw).
RO7019009 acts mechanistically after initial C99 binding.
RO7019009 reduces both pathogenic species and concomitantly increasing the short Aβ37 and Aβ38, of which the latter has been associated with delayed AD progression.
