RO7185876 (RO 7185876) is a potent and selective gamma secretase modulator (GSM) with IC50 Aβ42 of 2 nM.
RO7185876 demonstrates an excellent in vitro and in vivo DMPK profile, does not inhibit the Notch pathway,
RO7185876 reduced the amount of the large peptides Aβ42 and Aβ40 prone to aggregate and increases the fractions of smaller peptides Aβ37 and Aβ38 while leaving the total amount of Aβ unchanged.
RO7185876 demonstrated Aβ42 reduction effect in double transgenic mice model APP-Sweddish (free in vivo IC50 Aβ42=2.5 nM).
