RU-521 (RU.521, RU-320521) is a potent, selective cyclic GMP-AMP synthase (cGAS) inhibitor with Kd of 36.2 nM.
RU-521 inhibits cGAS-mediated interferon upregulation in cellular assays, with no effect on inflammatory pathways independent of cGAS.
RU-521 suppresses the chronically elevated levels of type I interferon in primary macrophages from mice model of AGS (Aicardi-Goutières syndrome).
