S3I-1757 is a small molecule STAT3-STAT3 dimerization inhibitor with IC50 of 13.5 uM in fluorescence polarization (FP) assays, interacts with Y-705 binding site in the SH2 domain.
S3I-1757 disrupts intracellular STAT3-STAT3 dimerization and STAT3-EGFR binding in intact cells.
S3I-1757 (50-200 uM) decreases phosphotyrosine STAT3 levels in the nucleus of human MDA-MB-468 breast cancer cells, inhibits STAT3-DNA binding.
S3I-1757 (50-200 uM, 18h) inhibits STAT3-dependent transcriptional activity and decreases the expression of STAT3-regulated genes in MDA-MB-468 cells.
S3I-1757 inhibits anchorage-dependent proliferation selectively, and inhibits migration and invasion selectively in human cancers that harbor high levels of P-STAT3.
