SALAD1 is a cell-permeable ultrabright RNA-activated fluorophore, facilitates improved in-cell confocal RNA imaging, binds to Mango II in a distinctive manner.
SALAD1 exhibitsa 3.5-fold improved brightness relative to TO1–biotin, which is typically used in cellular imaging with Mango aptamers.
SALAD1 (KDapp =0.69 ±0.1 nM) binds 7.5-fold more tightly to Mango II compared to TO (KDapp =5.9 ±1.4 nM).
SALAD1 exhibits a binding preference to its target RNA (Mango II) over the other three Mango aptamers (7-, 20- and 16-fold tighter than Mango I, III and IV.
