SDT-011 (SDT011) is a small molecule that effectively blocks the binding of anti-EGFR monoclonal antibodies to EGFR, blocks cetuximab-EGFR binding by 62% at 10 uM, binds to EGFR with Kd of 1.7 nM in MST assays.
SDT-011 does not bind cetuximab, interacts with the same residues on EGFR found to be important for the binding of EGFR mAbs cetuximab and panitumumab.
SDT-011 improves cell proliferation and EGFR activation in cell lines.
SDT-011 reactivates EGFR in ex vivo full human skin and in A431 injected mice.
SDT-011 can be delivered via a slow-release system derived from biodegradable nanoparticles that penetrate the hair follicles and sebaceous glands, within which EGFR is highly expressed.
