SGC-CK1γ-1 is the first chemical probe inhibitor of casein kinase 1γ (CK1γ) subfamily (CK1γ1, CK1γ2, and CK1γ3) with IC50 of 180,180 and 230 nM in NanoBRET assays respectively, inhibits both WNT signaling and human cytomegalovirus replication.
SGC-CK1γ-1 demonstrates a cellular IC50 of <1 μM, a biochemical IC50 <100 nM, good aqueous solubility in DLS, and excellent kinome-wide selectivity.
SGC-CK1γ-1 potently inhibits CK1γ1 / CK1γ2 / CK1γ3 with IC50 of 28/ 7/ 958 nM in enzymatic assays.
SGC-CK1γ-1 (5 uM) induces significant loss of LRP6 phosphorylation at T1479 in HEK293 cells, does not impact cell viability.
SGC-CK1γ-1 elicited anti-HCMV replication effects with no noticeable impact on cell viability when used at 10 µM.
