Biological Activity

SIS17 is a selective, cell-active HDAC11 inhibitor with IC50 of 0.83 uM.
SIS17 displays no inhibitory effect (IC50>100 uM) against Class I (HDAC1 and HDAC8), Class II (HDAC4), and the NAD-dependent Class III (sirtuins, SIRT1, SIRT2, SIRT3, and SIRT6).
SIS17 inhibits the demyristoylation of a myristoyl-SHMT2 peptide by HDAC11 in vitro with IC50 of 270 nM, inhibits HDAC11-mediated SHMT2 defatty-acylation in cells.
