Biological Activity
SU0547 is a specific small-molecule activator of DNA repair enzyme SMUG1 (Single-Strand-Selective Monofunctional Uraci-DNA Glycosylase 1) with AC50 of 4.3 uM, activates SMUG1 by more than 40-fold in vitro.
SU0547 activates SMUG1 by as much as 350% at 100 nM in vitro.
SU0547 exhibits minimal inhibition on Src family (Src, Lck, Abl, Csk, and Yes1) and clinically relevant kinases (EGFR, HER2, BRAF, RET, and FLT3).
SU0547 (3-5 uM) significantly reduces the cytotoxicity of 5-hmdU and decreases levels of double-strand breaks induced by the damaged nucleoside.
SU0547 is a useful tool to study the mechanisms of 5-hmdU toxicity and the potentially beneficial effects of suppressing damage to pyrimidines in cellular DNA.
