Savirin (S. aureus virulence inhibitor) is a small molecule inhibitor inhibiting agr quorum sensing in S. aureus by blocking the transcriptional function of AgrA.
Savirin inhibits RNA III levels in S. aureus, but not S. epidermidis, without affecting agr-independent growth.
Savirin (5 ug/mL) inhibited agr::P3 activation in reporter strains of all agr alleles, inhibits both AIP1-induced RNAIII and RNAIII produced at a longer time point without addition of exogenous AIP1 with no effect on exponential phase growth.
Savirin inhibits agr-regulated gene transcription and secreted virulence factor production.
Savirin inhibits AgrA-dependent transcription in clinical isolates.
Savirin treatment in vivo attenuates tissue injury and promotes agr but not Δagr bacterial clearance, promotes agr-dependent host defense in vivo and in vitro.
