PTSP is a specific and potent inhibitor of Vibrio LuxR/HapR proteins and LuxR homolog SmcR in Vibrio vulnificus, inhibits quorum sensing in multiple strains of V. vulnificus, V. parahaemolyticus, and V. campbellii.
PTSP significantly reduces protease activity in V. campbellii, V. coralliilyticus, and V. vulnificus, but not in V. cholerae, with IC50 of 6.8 nM, 78.1 nM and 18.3 nM for V. vulnificus strains ATCC 27562, CMPC6 and YJ016.
PTSP has a lower dissociation constant (Kd) for SmcR (0.23 μM) compared to LuxR (0.51 μM).
PTSP binds to SmcR drives allosteric unfolding of the N-terminal DNA-binding domain, induces it's specifically degradation by the ClpAP protease.
