Biological Activity

TH8321 (TH-8321) is a potent, selective NUDT15 inhibitor with IC50 of 35 nM.
TH8321 demonstrated satisfactory selectivity among a panel of related proteins, including NUDT22, NUDT18, NUDT12, NUDT9, NUDT5, NUDT2, MTH1, ITPase, dCTPase, and dUTPase.
TH8321 potently engaged and inhibited cellular NUDT15, TH8321 (5 uM) substantially potentiated the anti-HCMV efficacy of ganciclovir in HCMV infected MRC-5 cells, recapitulating the effect of NUDT15 knockdown.
