Biological Activity

TJ0113 acid is a UMI-77 derivative and mitophagy inducer, targets Mcl-1 binding to LC3A, selectively activates the PINK1-Parkin signaling cascade and enhances mitophagy flux in vitro and in vivo.
TJ0113 effectively induces the production of mitochondrial autophagosomes (APP) in hepatocytes.
TJ0113 induces mitophagy and ameliorates mitochondrial damage in hepatocytes of MASH mice.
TJ0113-induced mitophagy is dependent on the PINK1/Parkin pathway.
TJ0113 reduced body weight, liver weight, insulin resistance, glucose tolerance and liver function changes in MASH mice, reduces serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels in HFD-fed mice.
TJ0113 (3 mg/kg and 7 mg/kg, gavage) attenuates liver fibrosis (LF) in MASH mice.
TJ0113 inhibits the activation of NLRP3 inflammatory vesicles and ameliorates the inflammatory response in MASH mice.
TJ0113 induces autophagy in LX-2 cells.
TJ0113 (1 uM) inhibits TGF-β1 (10 ng/mL)-induced fibrotic response through activation of mitophagy in LX-2 cells, significantly reduces ROS production in LX-2 cells.
