Cadisegliatin (TTP399) is a hepatoselective, oral Glucokinase (GK) activator with EC50 of 304 nM and 762 nM for human GK at high glucose concentration (15 mM) and normoglycemia (5 mM), respectively.
TTP399 increases GK activity in the liver without affecting the physiological role of GK (active when plasma glucose concentrations are high and inactive at low/normal plasma glucose concentration).
TTP399 increases glucose metabolism in rat hepatocytes with EC50 2.39 and 2.64 uM in the presence of 15 mM glucose for lactate production and glycogen content, respectively.
TTP399 improves glycemic control in both animal models of type 2 diabetes and patients with type 2 diabetes.
