Cyclic-14-KI (CP14) is a specific, noncompetitive and allosteric antagonist of TrkC truncated isoform TrkC.T1, selectively antagonize TrkC, reduces CP14 reduces the affinity of NT3 binds to TrkC.T1 with Kd of 11.5 nM.
Cyclic-14-KI (CP14) does not inhibit TrkC-FL/NT3 and does not affect the binding of PTPs to either isoform.
Cyclic-14-KI (CP14) blocks up-regulation of TNF-α mRNA, does not have any effect on receptor family members TrkA or TrkB that are activated by their respective ligands NGF or BDNF.
Cyclic-14-KI (CP14) decreases TNF-α protein and cell death in retinal explants of RHOP347S mice.
Cyclic-14-KI (CP14) decrease photoreceptor cell death in the RHOP347S mice retina in vivo.
