Biological Activity
TrmD-IN-4 is a potent, covalent inhibitor of bacterial tRNA (m1G37) methyltransferase (TrmD) with IC50 of 4.43 uM, with minimal inhibition of human Trm5, shows selective growth inhibition in bacteria expressing TrmD.
TrmD-IN-4 displays potent activity against multiple Staphylococcus aureus strains, including a methicillin-sensitive strain (MSSA), methicillin-resistant strains (MRSA), and an enterotoxin producer strain (MIC = 1.56–6.25 μM) and moderate activity against additional Gram-negative organisms, including Haemophilus influenzae and Vibrio cholerae (MIC = 50 μM).
