Tuvusertib (M1774) is a potent, selective, orally bioavailable ATR protein kinase inhibitor, shows anticancer activity against cance cell with expression the ATR inhibition sensitizing mixed lineage leukemia fusion protein. Tuvusertib (M1774) shows antitumor activity as monotherapy in preclinical models with DDR pathway gene mutations, including an ATM mut nonsmall-cell lung cancer (NSCLC) xenograft model and an ARID1A mut gastric cancer xenograft model. Tuvusertib (M1774) also shows antitumor activity as monotherapy and in combination with the PARP inhibitor niraparib in xenograft models of BRCA mut high-grade serous ovarian cancer.
