UCB-9260 is a specific, orally active compound, inhibits tumour necrosis factor (TNF) signaling by stabilising an asymmetric form of the trimer, binds to TNF with Kd of 13 nM.
UCB-9260 is selective for TNF over other superfamily members.
UCB-9260 inhibited TNF-dependent cytotoxicity with a geometric mean IC50 of 116 nM, and a geometric mean IC50 of 120 nM, using human and mouse TNF, respectively.
UCB-9260 inhibited NF-κB with a geometric mean IC50 of 202 nM after TNF stimulation in HEK 293 cell-line.
UCB-9260 inhibited TNF-driven functional effects in vivo, showed a significant reduction of the clinical score in LPS challenged collagen antibody induced arthritis (CAIA) model.
