Biological Activity

UNC10415667 is a potent, benzaldehyde containing NSD2 degrader with DC50 of 460 nM in NSD2 HiBit cells, recapitulates SCF-FBXO22-dependent ubiquitination and potent degradation of NSD2.
UNC10415667 is a bout 2-fold more potent than UNC8732 and UNC10088 in the same assay.
UNC10415667 is dependent on the presence of Cys326 in FBXO22.
UNC10415667 (10 µM for 6 hrs) completely degraded both the long and short form of NSD2 in HEK293T cells.
