VU591 is a potent, selective renal outer medullary potassium channel (ROMK, Kir1.1) inhibitor with IC50 of 0.24 uM in T1+ flux assays.
VU591 has no effect on Kir7.1 at 10 uM, does not inhibit Kir2.1, Kir2.3 and Kir4.1, 150-fold selectivity over Kir6.2/SUR1.
VU591 blocks the intracellular pore of the channel and is a useful probe to explore the viability of ROMK as a diuretic target.
